January 24, 2017 Written by Catharine Paddock PhD The researchers recently discovered that dopamine cells in the brain area affected by Parkinson's disease are unable to protect against aging-related damage to the DNA of their mitochondria. Researchers in Norway suggest that an answer to what causes Parkinson's disease may lie in the mitochondria - the tiny powerhouses inside cells - of dopamine-producing cells. They found that dopamine cells in diseased brains are less able to protect against aging-related damage in their mitochondrial DNA than cells in healthy brains. Study leader Dr. Charalampos Tzoulis - a neurologist at the University of Bergen and Haukeland University Hospital, both in Norway - and colleagues hope that the discovery will lead to new treatments for Parkinson's disease. They report the findings in the journal Nature Communications. Parkinson's is a progressive, brainwasting disease that affects movement and can manifest as a range of symptoms, including: muscle rigidity; speech problems; tremors in the hands, limbs, jaw, and face; and impaired posture, gait, and balance. Despite decades of research, the exact causes of Parkinson's disease remain a mystery. Experts generally agree that a combination of genetic and environmental factors - both of which vary from person to person - are involved. An important known risk factor is age. The estimated risk of developing Parkinson's disease is 2-4 percent for people aged 60 and over, compared with 1-2 percent in the general population. The disease mainly affects dopamine-producing neurons or nerve cells in a brain structure known as the substantia nigra. As the disease progresses, these cells malfunction and die, depleting levels of dopamine, a chemical messenger that plays an important role in controlling movement. Many scientists believe that a better understanding of the mechanisms behind the destruction of dopamine cells could lead to treatments that stop or even reverse Parkinson's disease. For their study, Dr. Tzoulis and colleagues focused on the mitochondria of dopamine-producing cells. Mitochondria are tiny compartments inside cells that act as powerhouses; they digest nutrients and produce energy-rich molecules for the cell. Mitochondria have their own DNA, separate from the DNA in the cell nucleus, that contains code for building the powerhouses. Dr. Tzoulis explains: "It is known that the DNA of mitochondria is damaged during aging, causing failure in the power generators, lack of energy, and disease." The researchers conducted a detailed investigation of mitochondrial DNA in the brains of healthy older people and individuals with Parkinson's disease. They found that the dopamine-producing cells in the substantia nigra of the healthy brains were able to protect against aging-induced damage to their mitochondrial DNA by producing more healthy DNA. This replenishment process was much weaker in the diseased brains, resulting in a gradual loss of healthy DNA in the mitochondria of their dopamine cells. Dr. Tzoulis says he believes that the discovery reveals an important mechanism that normally defends the brain against aging-induced damage. This mechanism appears to malfunction in people who develop Parkinson's disease, leaving their brains more susceptible to the effects of aging. He concludes: "There is generally very little knowledge about the mechanisms causing Parkinson's disease. June 13, 2018 The link is thought to be due to genes that predispose people to the conditions GETTY IMAGES WEDNESDAY, June 13, 2018 (HealthDay News) -- Men and women with type 2 diabetes may face a significantly higher risk of developing Parkinson's disease later in life, new British research suggests. The finding of a link followed the tracking of Parkinson's diagnoses among millions of diabetic and non-diabetic patients who use the National Health Service in England. Study author Dr. Thomas Warner said that, after accounting for conditions that might mimic Parkinson's, the research showed that those with type 2 diabetes had a 32 percent greater risk of later developing the progressively debilitating neurological disorder. Escalated risk was even more dramatic among younger diabetes patients, aged 25 to 44, who were found to face a fourfold greater likelihood of eventually developing Parkinson's, according to the report. And adults with diabetes who had already developed diabetes-related health complications -- including damage to the retina, kidneys or nerves -- faced a 49 percent hike in their Parkinson's risk. Nevertheless, Warner stressed, "it is vital to realize that, overall, the majority of diabetic patients do not develop Parkinson's disease." Although the study could not prove a cause-and-effect relationship, Warner cited two possible reasons for a link between the two diseases. First, he said, there could be "shared genetic predisposition to develop [both] type 2 diabetes and Parkinson's." And then "there may be shared pathways in leading to development of diabetes and Parkinson's," he added. While the exact nature of such a connection remains unclear, Warner suggested it might involve the insulin production and glucose control problems that characterize diabetes. "Unlike most tissues in the body, brain cells are almost totally reliant on glucose as a source of energy," Warner noted. "So if there is a problem in how insulin controls the use of glucose by cells, this may affect certain groups of brain cells selectively." Warner is a professor of clinical neurology with the University College London Institute of Neurology, as well as the Queen Square Brain Bank for Neurological Disorder, both in London. He and his colleagues published their findings online June 13 in the journal Neurology. For their study, the investigators used data from the English "Hospital Episode Statistics" database to identify 2 million British patients newly diagnosed with diabetes from 1999 through 2011. This group was then stacked up against 6 million British patients who had initially sought care during the same time frame for non-diabetes related issues, such as sprains, varicose veins, appendectomies or hip replacements. The researchers found that just over 14,000 of the 2 million in the diabetes group were later diagnosed with Parkinson's, compared with about 21,000 of the 6 million others. That translated into a more than 30 percent greater risk for Parkinson's among those with diabetes, the researchers said. Among diabetic patients 25 to 44 years old, 58 of over 130,700 people developed Parkinson's, compared with 280 out of nearly 2.6 million similarly aged non-diabetics. That translated into a fourfold greater Parkinson's risk among those with diabetes, the researchers said. Drug regimens and smoking histories were not considered in the current analysis; nor were patients seeking diabetes care outside a hospital setting. Dr. Michael Okun, medical director of the National Parkinson's Foundation, called the findings "not surprising, as the collective evidence from multiple studies has been converging on the idea of some link or association between Parkinson's and diabetes." According to Okun, "There are many potential explanations for a link between the two diseases, but in younger patients, genetics likely plays a key role. In older patients, the degenerative process itself may disrupt brain-driven endocrine pathways, including those related to insulin and to sugar management." Okun, who also serves as the chair of neurology at the University of Florida in Gainesville, said, "At this point the mechanisms are unknown, and will require careful research." He said it's "important to stress that although diabetes drugs are currently candidates to treat or prevent Parkinson's disease, we are not recommending this approach until more compelling data is available." Researchers at Stanford University have been giving older mice injections of blood from younger ones. Remarkably - and as if taken from the pages of a vampire novel - the new blood turned back the clock on the older mice's cognitive declines. Now, one of those researchers has co-founded a new company that is set to try giving older humans with Parkinson's injections of certain components from younger people's blood. Ultimately, the firm aims to develop a synthetic version of whichever of the thousands of possible components of blood has this fountain-of-youth effect, as the supply of young people willing to donate their blood is very much limited. Researchers at a biotech firm co-founded by a Stanford scientist are giving Parkinson's patients transfusions of blood components taken from young donors in a controversial trial As the body ages, it undergoes broad-sweeping changes. That includes changes to the blood. Though the shift in our blood supply is subtle, it may have serious implications for our overall heath and innate ability to fight other age-related diseases. With age, the body's volume of water begins to decline and, with it, the volume of blood coursing through our veins. Meanwhile, the bone marrow becomes a less efficient producer of red blood cells. Red blood cell count is considered an important indicator of aging, and their decline has even been called a 'signal' for death. White blood cell count, too, falls slightly with age. These cells contain crucial white blood cells, which fight disease and infection. Subtle though these known changes are, evidence suggests that some ingredient or ingredients in younger blood are effective combatants of aging. The older mice's cognitive declines were reversed, they developed new brain cells, and performed just as well on tests of their cognitive functioning as the young ones did. Next, the team is taking a two-pronged approach to developing a therapy. They are testing the effects of each component protein, one-by-one, a tedious but key process to figuring out which has or have the healing properties. Simultaneously, Alkahest, the biotech firm that Dr Wyss-Coray co-founded, is beginning testing on 90 human patients with Parkinson's disease to see if it young (human) blood could have the same protective effects on their brains. The first patient was given their first injection on December 4, 2018. If the trial shows promise, and the parallel study successfully identifies the key part of blood, the research team may have the basis for making a synthetic. About 6.8 million people donate blood every year in the US, and less than 38 percent of the population is even eligible to donate blood or platelets. In previous tests, the researchers transfused blood from 18- to 30-year-old humans to older ones (mostly in their 70s and 80s). This age group is the least likely to donate blood, according to an analysis conducted last year. And even if there were more donors, the transfusion of blood from young donors to older ones has already raised ethical questions - and more than a few eyebrows in the past.
